ABSTRACT

OBJECTIVE To study the imp rovement effects of total flavonoids of Psidium guajava leaves on myocardial hypertrophy in hypertensive model rats. METHODS Ten rats were randomly selected from 60 healthy SD rats as the normal group ; other 50 rats established hypertensive model ,and 44 rats with successful modeling were randomly divided into model group , anisomycin group [p38 mitogen-activated protein kinase (p38 MAPK)activator,1 mg/kg],total flavonoids of P. guajava leaves+ anisomycin group (200 mg/kg total flavonoids+ 1 mg/kg anisomycin )and total flavonoids of P. guajava leaves group (200 mg/kg) by random volume mass ranking method ,with 11 rats in each group. Rats in normal group and model group were given 3% hydroxymethylcellulose sodium solution ,and other groups were given relevant solution intragastrically ,once a day ,for consecutive 6 weeks. Blood pressure (systolic blood pressure ,diastolic blood pressure ,mean arterial pressure ),cardiac index and left ventricular index were measured. The levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and IL- 6 in myocardial tissue were detected. The pathomorphological changes of myocardial tissue were observed. The expression of p 38 MAPK, phosphorylated p 38 MAPK (p-p38 MAPK),extracellular regulated protein kinase 1/2 (ERK1/2),phosphorylated ERK 1/2 (p-ERK1/2),c-Jun N-terminal kinase (JNK)and phosphorylated JNK (p-JNK)in myocardial tissue were detected. RESULTS Compared with normal group ,the systolic blood pressure ,diastolic blood pressure ,mean arterial pressure ,cardiac index ,left ventricular index as well as the levels of TNF-α,IL-1β and IL-6 and protein expression of p-p 38 MAPK,p-ERK1/2 and p-JNK in myocardial tissue were increased significantly in anisomycin group and model group (P<0.05);it was also found that hypertrophy of cardiomyocytes ,disorder of myocardial fibers ,looseness,edema and proliferation of connective tissue between myocardial fibers,increased infiltration of inflammatory cells ,etc. Compared with anisomycin group and model group ,the le vels of above indexes in total flavonoids of P. guajava leaves+ anisomycin group and total flavonoids of P. guajava leaves group were decreased significantly (P<0.05); cardiomyocytes were 163.com slightly larger and arranged reasonably ;the degree of myocardial hypertrophy,looseness,edema and proliferation of connective tissue were relieved ,and the improvement effect of total flavonoids of P. guajava leaves group was more significant (P<0.05). CONCLUSIONS The total flavonoids of P. guajava leaves can reduce blood pressure and improve myocardial hypertrophy in hypertensive model rats. Its mechanism may be related to the inhibition of p38 MAPK signal pathway activity and the expression of inflammatory factors.

ABSTRACT

Objective@#To investigate the clinical significance of the monocyte count/high-density lipoprotein cholesterol ratio(MHR)in evaluating imperfect ST-segment resolution in elderly patients with acute ST-elevation myocardial infarction(STEMI)after percutaneous coronary intervention(PCI).@*Methods@#This was a retrospective cohort study.A total of 274 elderly patients with STEMI underwent PCI in our hospital from December 2015 to December 2018 were enrolled.Based on the extent of the ST-segment resolution of the postoperative electrocardiogram, patients were divided into an imperfect ST-segment resolution group(observation group, n=79)and a favorable ST-segment resolution group(control group, n=195). General clinical data were compared between the two groups, and logistic regression equation was used to analyze the association of MHR with ST-segment resolution.Receiver operating characteristic(ROC)curve was performed to assess the predictive value of MHR for imperfect ST-segment resolution.@*Results@#Compared with patients in the control group, patients in the observation group were associated with a significantly higher proportion of anterior wall myocardial infarction and heart failure(≥Killip 2), A longer duration of chest pain to balloon expansion, higher levels of creatine kinase isoenzyme, N-terminal pro-brain natriuretic peptide, hypersensitive C-reactive protein, blood sugar, blood uric acid, fibrinogen, triglyceride and mononuclear cell count, and lower levels of high density lipoprotein cholesterol and lymphocyte count(all P<0.05). Meanwhile, there was a significant difference in MHR between the observation group and the control group [(0.75±0.22)vs.(0.48±0.19), t=9.831, P=0.001]. Multivariate Logistic regression analysis showed that MHR was an independent risk factor for imperfect ST-segment resolution(OR=1.950, 95%CI: 1.646-5.430, P=0.003)and ROC curve showed the threshold value of MHR at 0.67, the area under the curve at 0.867, the sensitivity at 79.72%, and the specificity at 79.61%.@*Conclusions@#MHR may be an independent risk factor and a good predictive index for imperfect ST-segment resolution in elderly patients with STEMI after PCI.

ABSTRACT

Objective: To evaluate the relationship between serum mRNA level of heparin binding epidermal growth factor (HB-EGF) and acute coronary syndrome (ACS) occurrence. Methods: Our research included in 2 groups: ACS group,n=50 patients and Control group,n=100 normal subjects. Serum HB-EGF mRNA level was examined by RT-PCR and the relationship between HB-EGF mRNA and ACS occurrence was assessed by Logistic regression analysis. Results: Compared with Control group, the serum HB-EGF mRNA level of ACS group was higher (0.22±0.73) vs (0.46±0.14),P<0.05. With adjusted meaningful factors of hypertension, smoking, TG, TC, LDL-C, HDL-C and BMI by single factor analysis, multi Logistic regression analysis indicated that serum HB-EGF mRNA was related to ACS occurrence (OR=5.813, 95% CI 2.342-14.426,P<0.001) which meant that upon 0.1 grey value of HB-EGF mRNA elevation, the risk of ACS occurrence may increase 4.813 folds accordingly. Conclusion: Serum HB-EGF mRNA level was related to ACS occurrence.

ABSTRACT

BACKGROUND:Bonemarrow mesenchymal stem cel transplantation can effectively improve decreased cardiac function caused by heart failure, but there is a lack of research about the effectinbone marrow mesenchymal stem cel transplantation on cardiac functioninheart failure induced by cardiomyopathies. OBJECTIVE:To explore the effect of bone marrow mesenchymal stem cel transplantation on cardiac function in patients with cardiomyopathiesaccompanied by heart failure. METHODS:Totaly 40 Sprague-Dawley rats were enroled, and bone marrow mesenchymal stem cels were isolatedfrom 10rats, and the remaining rats were equivalently randomized into normal, model and stem cel transplantation groups. Then rats in the model and stem cel transplantation groups were given intraperitoneal injection of hydrochloric acid doxorubicin to preparecardiomyopathy-inducedheart failure models. At 7 days after modeling, the stem cel transplantation group was treated with bone marrow mesenchymal stem celsthrough intravenous transplantation, and the model group was treated with equal amount of DMEM medium. Four weeks later, cardiac function of each rat was detected, and the cel survival and differentiation were observed by immunofluorescence method. RESULTS AND CONCLUSION:At 4 weeks after transplantation, compared with normal and stem cel transplantation groups, the left ventricular systolic pressure and maximumrise/fal rate of left ventricular pressurewere significantly decreased, buttheleft ventricular end diastolic pressure significantly increased in the model group (P 0.05).Highand dense fluorescence intensity was observed in the host myocardiumimmediatelyafter transplantation.Subsequently, the fluorescence intensityand density decreasedat 4 weeks, but the cel migration could be found, and some cels expressed cardiac troponin T. These results show thatintravenous transplantation ofbone marrow mesenchymal stem cels can improve cardiac function in rats withheart failure due tocardiomyopathies. Besides, the transplanted cels can survive in the host, and differentiate into cardiomyocyte-like cels.

ABSTRACT

Objective To study the changes of field potentials (FPs) during development and after long-term potentiation (LTP) induction in rat visual cortical slices. Methods Field potentials recorded from layer Ⅱ/Ⅲ induced by layer Ⅳ in the rat visual cortical slices were examined. The developing changes of FPs from postnatal days 8-13,15-21 and 29-35 were observed. Amplitudes, peak latencies and slopes of FPs before and after LTP induction were compared. Results Field potentials changed with the development, and the stimulating threshold at p15-21 was lower than that of the other two groups. The increase of FP amplitude was positively correlated with increase of its slope. The shortening of FP latency was positively correlated with the difference between changes of slope and amplitude. Conclusion Developing changes of field potentials reflect the development of visual cortex. The changes of FP slope follow the changes of its amplitude and latency, and thereby the slope is a proper parameter used as an index to evaluate LTP in visual cortex.

ABSTRACT

Objective To compare the different effects of different titanic parameters on the expression of long-term potentiation (LTP) and to determine the optimal patterns of tetanizing stimulation in hippocampal CA1 area in adult male rats. Methods Four tetanic parameters of 100Hz were used in the experiment. Tetanus 1 was a short burst of stimulus composed of 60 pulses, which were divided into 5 sets, each set including 4-pulses-bursts. The inter-burst intervals are lengthened gradually, 1s, 5s, 30s, 10min in sequence. Tetanus 2 was a long burst of stimulus composed of 300 pulses in all, each burst including 20 pulses. The inter-burst interval is 2s, and tetanus 3 is also a short burst of stimulus composed of 15 bursts, each burst including 4 pulses. The inter-burst interval is 2s and tetanus 4 is a single train of burst composed of 60 pulses. The field excitatory post-synaptic potentials (fEPSPs) in hippocampal CA1 area were evoked by stimulating the Schaffer collateral fibers in the stratum radiatum and the expressions of LTP induced by the four parameters were evaluated. Results The amplitude and incidence of LTP induced by tetanus 1 were higher than those induced by the other three parameters. There was no statistical significance between the responses induced by the other three parameters. Both tetanus 1 and tetanus 3 could lead to long-lasting-LTP (LL-LTP, over 3h). Conclusion A high frequency stimulus with 200ms-inter-burst interval (theta frequency) is better in the induction of LTP in hippocampal CA1 area. A short burst of stimulus with 100Hz can evoke the expression of LL-LTP in hippocampal CA1 area.

ABSTRACT

Objective To study the effects of gamma-aminobutyric acid (GABA) on the induction of long-term potentiation (LTP) in developing rat visual cortical slices. Methods Field potentials recorded from layer Ⅱ/Ⅲ induced by layer Ⅳ stimulation in rat visual cortical slices were examined. LTP was induced by tetanus in the slices on the postnatal 2, 3 and 5-week rats. The LTP incidences and the effects of GABA on them in different developmental stages were compared. Results LTP incidence at postnatal week 5 was significantly lower than that in other two groups. GABA A receptor antagonist picrotoxin could reverse this tendency, but had no effects on the LTP induction at the early postnatal age. Conclusion In visual cortex, GABA has inhibitory effects on the LTP induction only after the end of critical period and has no any facilitatory effects on the LTP induction before the developmental critical period.
